  • CK101302S Heat Stable Recombinant Human bFGF

CK101302S Heat Stable Recombinant Human bFGF (HSbFGF) is an E. coli-expressed, genetically engineered thermostable mutant derived from wild-type bFGF through directed engineering. After 4 days of incubation at 37°C, HSbFGF retains full activity (vs. wild-type bFGF dropping to 30%), enabling sustained maintenance of ESC/iPSC undifferentiated state without frequent medium replenishment. Suitable for long-term stem cell culture, MSC expansion, and neural stem cell expansion. Animal-free production system, lyophilized powder form, store at -20°C.

    Background

Basic fibroblast growth factor (bFGF, FGF-2) belongs to the fibroblast growth factor family. It specifically binds to tyrosine kinase receptors and activates the FGF/FGFR signaling pathway, subsequently transducing other classical pathways that influence cell proliferation, differentiation, apoptosis, and immune regulation. bFGF functions include: regulating cartilage metabolism via modulation of the JAK-STAT signaling pathway; acting as a mitogenic promoter to accelerate cell proliferation. bFGF is suitable for cell culture and is a key component for maintaining embryonic stem cells in an undifferentiated state during culture. Heat-stable bFGF (HSbFGF) is generated from the wild-type sequence through genetic engineering. Throughout the cell culture period, wild-type bFGF tends to aggregate and precipitate, leading to loss of activity, whereas HSbFGF remains stable. In thermal acceleration experiments, after 4 days of incubation at 37°C, wild-type bFGF activity decreased to 30%, while HSbFGF activity remained unchanged. Furthermore, HSbFGF exhibits biological functions in stem cell culture identical to those of wild-type bFGF. Based on its thermal stability and biological characteristics, HSbFGF is better suited for stem cell culture. This product is a recombinant human HSbFGF protein produced in an E. coli system, featuring high purity and freedom from animal-derived contamination.

    Directions for Use

This product is in lyophilized powder form. It is recommended to reconstitute with sterile water, PBS, or other suitable buffer to prepare a stock solution at 0.1–1.0 mg/mL.

After complete dissolution, further dilute to working concentration using culture medium according to experimental requirements. Aliquot for single-use storage and store at -20°C or below. Avoid repeated freeze-thaw cycles to maintain product activity and stability.

Optimal bFGF concentration may vary across different cell types and culture systems. Optimization and validation based on specific culture processes and experimental objectives are recommended to achieve the best culture results.

    Transport & Storage

    Ice pack transport.

Store at -20°C or below, protected from light, moisture-proof, and sealed. After reconstitution, aliquot and store at -20°C or below. Avoid repeated freeze-thaw cycles. Minimize product exposure to room temperature during handling to ensure product activity and stability.

    Applications

Recombinant Human bFGF (FGF-2) is a critical regulatory factor in stem cell culture. By binding to FGFR tyrosine kinase receptors and activating downstream signaling pathways, it maintains the undifferentiated state and self-renewal capacity of embryonic stem cells (ESC) and induced pluripotent stem cells (iPSC), while also regulating the proliferation and differentiation of mesenchymal stem cells (MSC) and neural stem cells.

HSbFGF, through genetic engineering, has significantly enhanced thermal stability. Its activity remains unchanged after 4 days of incubation at 37°C, solving the pain point of rapid inactivation and frequent replenishment of wild-type bFGF in culture systems, making it especially suitable for long-term stem cell culture.

    Widely used in:

    • ESC/iPSC pluripotency maintenance and long-term culture

    • Mesenchymal stem cell (MSC) expansion

    • Neural stem cell (NSC) expansion

    • Organoid culture (brain, intestinal, etc.)

    • Serum-free medium supplement

    • Cell proliferation and differentiation research

    • Tissue engineering and regenerative medicine

    • Biopharmaceutical R&D

    Precautions

    • This product is intended for research, process development, and biomanufacturing use only. Not for diagnostic, therapeutic, or clinical use in humans or animals.

    • Use the product as soon as possible after opening. Unused portions should be promptly sealed and stored.

    • Maintain aseptic technique throughout reconstitution and handling to avoid microbial contamination.

    • Aliquot for single-use storage to avoid repeated freeze-thaw cycles that may compromise product activity.

    • Optimal bFGF concentration may vary across different cell types and culture systems. Determine optimal usage conditions through preliminary experiments.

    • Read the product technical datasheet carefully before use and follow relevant experimental protocols.
