    Background

    CD16 is a low-affinity Fc receptor comprising two subtypes — FcγRIIIa (CD16a) and FcγRIIIb (CD16b) — that specifically bind the Fc region of IgG antibodies. These two highly homologous genes are expressed in different cell types and are widely distributed on the surface of natural killer cells (NK cells), neutrophils, polymorphonuclear leukocytes, monocytes, and macrophages. CD16a (also known as FcγRIIIα) is a medium-affinity polypeptide-anchored transmembrane glycoprotein primarily found on NK cells, macrophages, certain T cell subsets, immature thymocytes, and placental trophoblasts. It plays important roles in phagocytosis, enzyme secretion, inflammatory mediator release, antibody-dependent cellular cytotoxicity (ADCC), and immune complex clearance. CD16b (FcγRIIIb) is a GPI-anchored low-affinity receptor mainly expressed on neutrophils and eosinophils. Studies have shown that CD16 stimulation triggers ζ chain tyrosine phosphorylation in NK cells, accompanied by increased intracellular Ca²⁺ concentration and elevated IP3 levels, thereby activating cytokine synthesis and ADCC activity. This product is a high-purity humanized recombinant monoclonal antibody expressed in CHO cells. The formulation is PBS buffer, free of carrier proteins or other additives. The production process uses completely chemically defined serum-free medium and alkali-tolerant Protein A chromatography purification, ensuring freedom from serum, animal-derived components, and adventitious agents, significantly enhancing product safety, stability, and reliability for cell culture and diverse scientific research applications.

    Directions for Use

    This product is in liquid form at a concentration of 1 mg/mL in 1× PBS (pH 7.4).

    Thaw at 2–8°C or room temperature before use and mix gently. Further dilute to working concentration using culture medium or suitable buffer according to specific experimental requirements.

    For CAR-NK and NK cell activation experiments, optimization of antibody concentration based on cell density, culture system, and experimental objectives is recommended. Aliquot for single-use storage to avoid repeated freeze-thaw cycles.

    Applications

    Humanized anti-human CD16 monoclonal antibody (Anti-CD16 mAb) specifically targets the FcγRIII receptor (CD16a/CD16b). CD16 is a low-affinity receptor for the IgG Fc region, primarily expressed on the surface of NK cells, neutrophils, monocytes, and macrophages. Upon antibody binding to CD16, signal transduction is initiated through tyrosine phosphorylation of ITAM motifs on the ζ chain (or FcεRIγ chain), inducing intracellular Ca²⁺ elevation and IP3 generation, and triggering ADCC effector function and cytokine release (IFN-γ, TNF-α, etc.).
